    Mechanisms of STAT family mediated IDO regulation in dendritic cells

    Indoleamine 2, 3-dioxygenase (IDO) is known as an endogenous immunosuppressive enzyme which plays a significant role in the process of tumor. IDO is not only found in tumor cells but also detected in dendritic cell (DC) in tumor microenvironment, which participates in the formation of tumor immune tolerance through expressing IDO enzyme. Signal transducer and activator of transcription (STAT) is the main signal protein family which participates in the IDO transcriptional regulation of DC.It is necessary to detail the signaling pathway in regulating IDO expression, which will help us develop high specific andmore active IDO inhibitors and provide new options for anti-cancer targeted therapy.

    Toll-like receptor (TLR) is an important pattern recognition receptor (PRR) which participates in innate immunity and regulates adaptive immunity. TLR can be expressed in immune cells and malignant tumors recognize conservative molecular structure, mediate response of inflammation, tissue injury and repair, which plays an important role in the process of tumor. Research results about some molecules and signal pathways of TLR demonstrate that it can act anti or pro-tumor dual functions, which has an extensive prospects in prevention and treatment of tumor.

    Ecotropic viral integration site-1 (Evi-1) gene is first identified as a binding site for retrovirus in murine myeloid leukemia. The human homologue with the murine in leukemia oncogene Evi-1 is about 91% of nuclear sequence and 94% of amino acid. Evi-1 gene is high expressed in myeloid malignancies and other tumors. The main tumorigenic mechanisms of Evi-1 gene include inhibiting TGF-β signal path by depressing Smad3, adjusting cell cycle, promoting angiogenesis, inhibiting JNK, activating AP-1, and so on.

    PRDI-BF1 and RIZ homology domain containing proteins (PRDM) play a key role in cell differentiation and proliferation. Most members of the PRDM gene family are  tumor suppressor genes which involved in tumorigenesis and abnormal expression in a variety of tumors. Aberrant DNA methylation often silences these genes, which may occur in the early stage of tumor. Cancer can be reversed by demethylation, which provides a new way for cancer treatment.

    Epithelial-mesenchymal transition (EMT) not only can endow cells migration and invasion characteristics, but also can make tumor cells obtain self-renewal ability and have the characteristics of stem cells, which might result in cancer stem cell (CSC). There are the same molecular mechanism and microenvironment between EMT and CSC, which have great clinic significances for the diagnosis and treatment of the aggressive cancers. Moreover, many studies show that miR-200 could regulate EMT and CSC, participate in the tumor invasion and metastasi, and promote the research of targeted cancer therapy.

    Interleukin-10 (IL-10) is an immuno-regulatory cytokine, which is well known for its immunosuppressive function. IL-10 can act on a variety of immune cell subsets and play an immunosuppressive effect through a variety of ways. It plays an important role in the tumor immune escape. Recent studies show that IL-10 also has an immune-stimulating function, which is presumably a pluripotent cytokine. This paper will combine the latest literature to summary the positive and negative regulatory roles of IL-10, mediation ways in the tumor and the relationships with prognosis.

At present, studies show that Bmi-1 is highly expressed in a variety of solid tumors, which can be regarded as one of the markers of cancer stem cells and has a close relation to the cancer development and progression and radiation resistance. It will provide an important target gene for the treatment of malignant tumors.

Morphological observation and invasion-related protein detection can be used to evaluate the animal invasion model, which are widely used in basic and clinical research now.

    Regorafenib, an oral multi-kinase inhibitor, can inhabit a class of receptor tyrosine kinase, such as angiogenic, stromal, oncogenic and so on. Studies in vitro and clinical trials indicate that regorafenib has significant antitumor activity. The results of clinical trials are encouraging for the treatment of refractory solid tumors, especially for colorectal carcinoma.

    The use of adjuvant external-beam RT is well established in the postoperative treatment of glioblastoma multiforme (GBM). It is consensus that target volume should be determined based on the fusion images of MRI and CT, but the inclusion of peritumoural edematous is controversial. The vast majority of recurrences occur within 2 cm of the original tumor site or “in radiation field”. There is no inevitable relation between the degree of peritumoral edema and recurrence model. The clinical and pathological characteristics may be as predictive and prognostic factors for the treatment of GBM. Target volume delineation for GBM tend to individual, which can maintain known outcomes and reduce treatment toxicity.

    The research of esophageal target is a difficult spot in the precise radiotherapy and the target mobility has an important effect for the radiotherapy of esophageal cancer. By different breathing control techniques, controlling the patient's respiratory may narrow target mobility so as to improve the accuracy of radiotherapy. But the present studies of the movement range of esophageal cancer with the respiratory control technologies have not come to a consistent standard. The area shape and position change of esophageal cancer target is a key problem that needs to be solved in the esophageal precise radiotherapy.

The possible mechanisms include: iodine complex can inhibit breast cell proliferation and can induce apoptosis; thyroid hormone can affect breast cancer cell proliferation and apoptosis; the imbalance of thyroid hormone and estrogen hormone promotes breast cells cancerous; hypothyroidism may affect the proliferation and invasiveness of breast tumors; lymphocytic infiltration or abnormal local immune response in thyroid disease.

    Hedgehog (HH) signaling pathway is a classic signaling pathway which controls embryonic development. Recent studies show that HH signaling pathway plays an important role in the tumorigenesis and development of cancer. Breast cancer stem cell (BCSC) is closely associated with malignant tumor metastasis, relapse and treatment resistance. HH pathway plays crucial roles in maintaining the characters of BCSC, and will probably become a novel therapeutic target of breast cancers.

    Women diagnosed with breast cancer have a significantly higher risk of developing contralateral breast carcinoma. When the dignosis of “bilateral breast cancer (BBC)” is established, the challenges for the doctors are to determine contralateral breast carcinoma is metastasis or primary lesions. The pathological type, receptor status, biological behavior and the prognosis of bilateral breast cancer are the issues have to be considered. On the other hand, we discuss which type of breast cancer trend to be bilateral breast cancer, thus more effective follow-up and preventive procedures would be applied on the patients with bilateral breast cancer to improve their quality of life and the curative rate.

Researches about Fas/FasL protein expression and Fas/FasL-mediated apoptosis, immune escape mechanism and their roles in the pathogenesis, progression and prognosis of lung cancer are constantly emerging. Discussion about the roles of Fas/ FasL system in NSCLC could provide evidence for early diagnosis, prognosis prediction and new treatment of NSCLC.

The application of new techniques such as immunohistochemical method and reverse transcription polymerase chain reaction (RT-PCR) can study the lymphatic metastasis from the genetic and molecular level.  The application of molecular imaging and nanoparticles could provide new detection methods and therapies. The establishment of animal models offers a solid foundation for experimental study.

    Inhibition of resveratrol on proliferation of different breast cancer cell lines

    Objective  To investigate the effects of resveratrol (Res) on the proliferation, cell cycle and apoptosis of three breast cell lines DU4475, MDA-MB-231 and MCF-7 with different estrogen receptor (ER) expressions. Methods  Res was added to the drug treatment group at 6.25, 12.5, 25, 50, 100 and 200 μmol/L, respectively. Three observation periods at 24, 48 and 72 hours were set up respectively. MTT assay was used to detect the effects of Res on proliferation of breast cells. Cell cycle and apoptosis were analyzed by flow cytometry (FCM). The concentrations of drugs were 0, 12.5, 25, 50 μmol/L. The results were analysed by the statistical software SPSS17.0. Results  After Res intervention, the proliferations of three cell lines were inhibited to different extent. After 48 and 72 hours of Res, inhibitions of Res with different concentration were significant different (F=15.26, P﹤0.05). Inhibition rates of DU4475 and MDA-MB-231 with ER-negative were higer than that of MCF-7 with ER-positive. FCM results prompted that these three kinds of cells were blocked in S phase after 48 hours treatment of 12.5-50 μmol/L Res and the block percentages had significant difference (F=34.81, P﹤0.05). The percentages of S phase for DU4475 and MDA-MB-231 arresting were higher than that of MCF-7. For DU4475, the apoptotic and necrosis percentage were higher than that of MCF-7 at 100 μmol/L (t=16.082, P<0.05). Conclusion  Res can inhibit proliferation, induce cell cycle changing and apoptosis of DU4475, MDA-MB-231 and MCF-7 cells. The inhibitions of Res on DU4475 and MDA-MB-231 are better than that of MCF-7 with ER-positive.

    Expression and significance of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus

     Objective To investigate the expressions and significances of SIRT1 and p53 in endometrial carcinoma with diabetes mellitus. Methods Immunohistochemistry was carried out to examine the expressions of SIRT1 and p53 in 39 cases of endometrial carcinoma with diabetes mellitus and 40 cases of endometrial carcinoma without diabetes mellitus and 25 cases of normal endometrium, and the correlations between these indexs were analysed. Results The positive rate of SIRT1 and p53 in experimental group was 64.1%, 59.0% respectively, and 85.0%, 52.5% in control group. The positive rate of SIRT1 in experimental group was lower than control group (χ2=4.561, P=0.033). The positive rates of SIRT1 and p53 in control group were higher than normal endometrium (χ2=21.462, P<0.001; χ2=6.771, P=0.009). Expression of SIRT1 was positively correlated with p53 (r = 0.360,P =0.024). Conclusions Hyperglycemia deregulates SIRT1 in endometrial carcinoma. SIRT1 may promote tumorigenesis by silence of p53.
