摘要: 蛋白酶Nexin-1(PN-1)是Serpin类超家族的重要一员,在人类胰腺癌、乳腺癌、结肠癌、睾丸癌等多种恶性肿瘤细胞中异常表达,通过抑制多种蛋白酶、细胞外信号激酶(ERK)信号通路,参与肿瘤细胞的增生、分化、凋亡、血管新生和浸润,在肿瘤的发生发展及转移中起促进作用。特别是在前列腺癌中,PN1通过抑制尿激酶纤溶酶原激活剂(uPA)、Hh信号通路以及与基质金属蛋白酶(MMP)9的相互作用,在肿瘤的发生发展及转移中起抑制作用。

Abstract: Protease nexin-1 (PN-1) is one of the most important members of Serpin superfamily. The abnormal expressions are observed in many human cancers,  such as pancreatic cancer, breast cancer, colon cancer, testicular cancer and so on. PN1 participates in tumor cell proliferation, differentiation, apoptosis, angiogenesis and invasion by inhibiting multiple proteases and extracellular signal regulated kinase (ERK) pathways and promotes tumor occurrence, development and metastasis. In prostate cancer particularly, PN1 inhibits tumor occurrence, development and metastasis by inhibiting urokinase plasminogen activator (uPA), Hh signaling pathway and interacting with matrix metalloproteinase (MMP)-9.
