铁死亡作用机制及其与肿瘤的关系

摘要: 铁死亡是近年来发现的一种新型细胞死亡方式,是铁依赖的脂质过氧化积累介导的细胞死亡,其在形态及生物化学上不同于凋亡、坏死等细胞死亡方式。铁死亡发生过程中伴随着谷胱甘肽的耗竭、谷胱甘肽过氧化物酶活性的下降以及大量脂质过氧化物的产生。研究发现,铁死亡可参与肿瘤的发生、进展,激活铁死亡通路中不同的调节位点可促进肿瘤细胞的死亡。因此,明确铁死亡通路及其调节肿瘤发展的相关机制可能为临床治疗肿瘤提供新的思路。

Abstract: Ferroptosis is a new type of cell death discovered in recent years. This form of irondependent cell death is morphologically and biochemically distinct from other cell death modalities, including apoptosis and necrosis. This process is accompanied by the depletion of glutathione, a decrease in glutathione peroxidase activity and the accumulation of lipid peroxidation products. Studies have found that ferroptosis can be involved in the occurrence and progression of tumor by activating different regulatory sites in the signaling pathways, which can promote tumor cell death. Therefore, to clarify ferroptosis pathway and its related mechanisms regulating tumor development may provide new ideas for clinical treatment of tumors.
