MGMT和Ki67及P53蛋白在脑胶质瘤中表达及意义

Expression and clinical significance of MGMT, Ki 67 and P53 in human gliomas

摘要: 目的探讨DNA修复酶O6甲基鸟嘌呤DNA甲基转移酶(MGMT)、细胞增殖相关核蛋白Ki67和P53蛋白在脑胶质瘤中的表达情况与分级的关系及其临床意义。方法采用免疫组织化学SP法检测61例脑胶质瘤标本和16例外伤内减压正常脑组织标本MGMT、Ki67和P53蛋白的表达情况。结果MGMT(19.67%∶0, χ2=3.729,P=0.062)、Ki67(39.34%∶0, χ2=5.722,P=0.016)和P53蛋白(27.87%∶0, χ2=9.146,P=0.002)在胶质瘤中的表达和正常脑组织中的表达差异均有统计学意义,且Ki67在高级别胶质瘤(Ⅲ~Ⅳ级)与低级别胶质瘤(Ⅰ~Ⅱ级)差异有统计学意义(14∶10,χ2=11.718,P=0.001)。MGMT、P53未发现明显差异。结论MGMT蛋白表达可作为胶质瘤检测的生物学标志物;Ki67蛋白与病理分级存在正相关,可作为脑胶质瘤病理分级的参考指标;P53蛋白可能为胶质瘤治疗的潜在靶点。

Abstract: ObjectiveTo detect the expression of  DNA repair enzyme O6methylguanine DNA methyltransferase (MGMT), cell proliferationrelated nuclear protein (Ki67) and P53 protein in gliomas, and investigate the relationship and clinical significance among them and gloma grade. Methods61 cases of brain glioma specimes and 16 cases of internal decompression of brain trauma were used to detect the expression of MGMT, Ki67 and P53 by using immunohistochemical SP method. ResultsMGMT protein expression (19.67%∶0, χ2=3.729,P=0.062), Ki67 protein expression (39.34%∶0, χ2=5.722,P=0.016) and P53 protein expression (27.87%∶0, χ2=9.146,P=0.002) showed significant differences in gliomas compared to normal brain tissues, the expression of Ki67 was significantly higher in highgrade gliomas(ⅢⅣ) than that in lowgrade gliomas (ⅠⅡ) (14∶10, χ2=11.718,P=0.001). No significant difference was found between the MGMT and P53.ConclusionMGMT protein can be used as a biomarker in gliomas detection; Ki67 has a positive correlation with tumor grade, and can be used as a reference indicator of pathological grade; P53 protein expression may be used as a potential target for the treatment of gliomas.
