HIF-1α、PDGF与VEGF、VEGFR在乳腺癌发病中的表达及意义

Expression and significance of HIF-1α, PDGF and VEGF/VEGFR-2 in breast cancer

摘要: 目的探讨缺氧诱导因子-1α(HIF-1α)、血小板衍生生长因子(PDGF)、促血管内皮生长因子(VEGF)血管内皮生长因子受体(VEGFR)信号通路效应与乳腺癌发病、转移的相关性。方法对61例乳腺癌病理标本及17例乳腺增生、纤维瘤标本进行组织形态学和HIF-1α、PDGF-A、VEGF-A、VEGFR-2免疫组织化学研究并进行比较。结果61例乳腺癌标本中,39例VEGF表达阳性,其中20例(51.3%)阳性表达区域超过肿瘤组织的10%; 42例VEGFR表达阳性,有22例(52.4%)阳性表达区域超过肿瘤组织的10%。在VEGF阳性标本中同时VEGFR-2表达阳性的有12例。HIF-1α表达阳性50例,PDGF表达阳性39例,分别有36例(72.0%)和23例(59.0%)阳性表达区域超过肿瘤组织的10%。VEGFR的表达与患者年龄(χ2=11.080,P=0.001)及淋巴结转移(χ2=4.699,P=0.046)密切相关,而VEGF(χ2=0.880,P=0.415)、HIF-1α(χ2=1.620,P=0.303)及PDGF(χ2=0.150,P=0.786)与患者年龄未见明显相关;VEGF(χ2=0.677,P=0.437)、HIF-1α(χ2=1.394,P=0.323)及PDGF(χ2=1.841,P=0.192)与患者淋巴结转移亦未见明显相关。乳腺增生及增生性纤维瘤患者部分标本中亦有HIF-1α、PDGF-A、VEGF-A和VEGFR-2阳性表达,但均明显低于乳腺癌组织中的表达。结论HIF-1α、PDGF、VEGF-A、VEGFR-2在乳腺癌组织中均高表达,而VEGFR-2与患者发病年龄及淋巴结转移关系密切,提示上述蛋白可作为诊断、预后和靶向治疗的辅助标志物。

Abstract: ObjectiveTo discuss the relationship among hypoxiainducible factor1α (HIF1α), platelet derived growth factor (PDGF) and vascular endothelial growth factor (VEGF)/VEGFR signaling pathway in breast cancer occurring and metastasis in the early stage. MethodsThe clinical and pathological features and the expression of HIF1α, PDGFA, VEGFA and VEGFR2 were assayed by histomorphology and immunohistochemical staining on tissue specimens from 61 patients of breast cancer and healthy breast tissues healthy breast tissue from 17 patients of benign breast disease. ResultsVEGF expression was observed in 39 out of 61 malignant breast tissues, more than distribution of positive staining 20 patients (51.3%) of which. VEGFR expression was found in 42 out of 61 lesions,. more than 10% distribution of positive staining 22 patients (52.4%) of which. Simultaneous expression of VEGF and VEGFR2 was found in 12 cases. HIF1α was expressed in 50 and PDGF in 39 out of 61 breast cancer patients, and in 36(72.0%) HIF1α positive specimens and 23 (59.0%) PDGF positive specimens the staining area was more than 10%. The expression of VEGFR was  correlated with patient′s age (χ2=11.080, P=0.001)and lymph node metastasis (χ2=4.699, P=0.046). VEGF, HIF1α and PDGF were not correlated with patient′s age (χ2=0.880,  P=0.415; χ2=1.620, P=0.303; χ2=0.150, P=0.786) and lymph node metastasis (χ2=0.677, P=0.437; χ2=1.394, P=0.323; χ2=1.841, P=0.192). ConclusionHIF1α, PDGF, VEGF and VEGFR2 express in breast tumor tissues at a high level, and VEGFR2 is correlated with patient′s age and lymph node metastasis. Abovementioned proteins can be used as markers for diagnosis, prognosis and targeted therapy.
