白细胞介素28B rs12979860 T/C基因多态性与肝细胞癌易感性关系的Meta分析

Association between polymorphism of interleukin28B rs12979860 T/C and susceptibility of hepatocellular carcinoma: a Metaanalysis

摘要: 目的 探讨白细胞介素28B(IL28B)rs12979860 T/C基因多态性与肝细胞癌(HCC)易感性的关联。方法 计算机检索PubMed、EMBase、中国知网数据库、中国生物医学文献数据库、维普数据库及万方数据库,检索时间截至2014年9月30日,收集有关IL28B rs12979860 T/C基因多态性与肝细胞癌易感性的病例对照研究。由2名研究者按照纳入和排除标准独立选择文献、提取资料,采用RevMan 5.2和Stata 12.0软件进行Meta分析。结果 最终纳入6个病例对照研究,包括1 138例患者和955例对照。各遗传模型Meta分析结果显示,IL28B rs12979860 T/C多态性与肝细胞癌易感性的关联性有统计学意义,与基因型CC及CT+CC比较,基因型TT可增加人群罹患肝细胞癌的风险(TT∶CC:OR=2.26,95%CI为1.40~3.64,Z=3.33,P=0.000 9;TT∶CT+CC:OR=1.90,95%CI为1.23~2.93,Z=2.89,P=0.004)。种族来源的亚组分析结果显示,在白种人群中,IL28B rs12979860 T/C多态性与肝细胞癌易感性存在显著相关性(TT∶CC:OR=2.06,95%CI为1.22~3.47,Z=2.70,P= 0.007;TT∶CT+CC:OR=1.71,95%CI为1.07~2.72,Z=2.23,P= 0.03)。结论 IL28B rs12979860 T/C基因多态性与肝细胞癌易感性相关,基因型TT可增加罹患肝细胞癌的风险。

Abstract: ObjectiveTo explore the correlation between polymorphism of interleukin28B (IL28B) rs12979860 T/C and susceptibility of hepatocellular carcinoma(HCC). MethodsAll eligible casecontrol studies published up to 20140930 were identified by searching PubMed, EMBase, CNKI, CBM, VIP and WanFang databases. Two reviews independently identified the literature according to inclusion and exclusion criteria. Metaanalysis was performed using Rev Man 5.2 and Stata 12.0 software. ResultsA total of 6 studies comprising 1 138 cases and 955 controls were finally included. Metaanalysis showed that IL28B rs12979860 T/C polymorphism was associated with the susceptibility of HCC. Compared with the genotype CC and CT+CC, genotype TT increased the risk of suffering from HCC(TT vs CC: OR=2.26, 95%CI: 1.403.64, Z=3.33, P= 0.000 9; TT vs CT+CC: OR=1.90, 95%CI: 1.232.93, Z=2.89, P= 0.004). In stratification analysis by ethnicity, we observed that the polymorphism of IL28B rs12979860 T/C was associated with the susceptibility of HCC among Caucasian populations (TT vs CC: OR=2.06, 95%CI: 1.223.47, Z=2.70, P=0.007; TT vs CT+CC: OR=1.71, 95%CI: 1.072.72, Z=2.23, P= 0.03). Conclusion The polymorphism of IL28B rs12979860 T/C is associated with the susceptibility of HCC, genotype TT may increase the susceptibility to HCC.
